ABSTRACT

In this paper, the name, origin, producing area, quality evaluation, harvesting and processing methods of Scrophulariae Radix used in the famous classical formulas were researched by consulting related ancient materia medica, medical books and prescription books. The results showed that the name of Scrophulariae Radix originated from its shape and color. Scrophularia ningpoensis, one of the eight flavors of Zhejiang, has been highly praised in the past as a genuine base, and its authentic production area is Zhejiang. Quality of Scrophulariae Radix is best if it has a dry body, thin skin, thick branches, firm physique, black inner color, net basal part of stem, and no fine whiskers. In ancient times, the origin processing was divided into two types, including sun-dried after steaming and directly sun-dried. While it mostly used a combination of drying and sweating in modern times. Before the Ming dynasty, the processing of Scrophulariae Radix was mainly steamed. Occasionally, the stir-frying appeared. In the Ming dynasty and after the Ming dynasty, wine processing emerged such as hot wine mixed roasting, wine washing, wine steaming, because the ancients believed that wine could enhance its efficacy and reduce its cold property. In the Qing dynasty, the method of steaming and baking appeared in the water and fire system. According to the 2020 edition of Chinese Pharmacopoeia, the processing of Scrophulariae Radix includes removing rhizome residues and impurities, washing, moistening, cutting into thin slices, and drying, or soaking slightly, steaming thoroughly, drying slightly, cutting into thin slices, and drying. Referring to the relevant national documents, it is suggested that raw products of Scrophulariae Radix should be used in Liangditang and Simiao Yong′antang.

ABSTRACT

In this paper, the name, varieties, raw materials and manufacturing technology of maltose in the famous classical formulas were researched by consulting the herbal medicines, medical books, prescription books and modern literature of past dynasties, which provided the basis for the development and utilization of formulas containing maltose. Through textual research, it can be seen that the name of maltose has been derived from its shape, texture, preparation method, raw materials and producing area. In ancient times, maltose was mainly divided into soft and hard types according to the texture. Those who are wet and soft as honey are called "syrup" or "jelly", while those who are hard and white are called "malt" and "sugar". In modern times, they are mostly called malt sugar, only jelly is used as medicine, and malt is mostly used as food. Throughout the ages, medicinal maltose were made of Oryza sativa var. glutinosa as the raw material and Hordeum vulgare as malt, prepared by fermenting, decocting and concentrating. The maltose made from other cereals such as Setaria italica var. germanica, Panicum miliaceum is slightly inferior in quality. The 2020 edition of Chinese Pharmacopoeia did not include maltose, but included malt sugar, a pharmaceutical excipient, which was a crystal powder with high purity. But maltose was included in the national food standard and the local processing specification. Based on the textual research results, it is suggested that malt syrup in GB/T 20883-2017 can be used as the reference for the development of formulas containing maltose, and O. sativa var. glutinosa, H. vulgare are clearly used as raw materials.

ABSTRACT

BACKGROUND@#Numerous studies have concentrated on high-dose radiation exposed accidentally or through therapy, and few involve low-dose occupational exposure, to investigate the correlation between low-dose ionizing radiation and changing hematological parameters among medical workers.@*METHODS@#Using a prospective cohort study design, we collected health examination reports and personal dose monitoring data from medical workers and used Poisson regression and restricted cubic spline models to assess the correlation between changing hematological parameters and cumulative radiation dose and determine the dose-response relationship.@*RESULTS@#We observed that changing platelet of 1265 medical workers followed up was statistically different among the cumulative dose groups (P = 0.010). Although the linear trend tested was not statistically significant (P@*CONCLUSION@#We concluded that although the exposure dose was below the limit, medical workers exposed to low-dose ionizing radiation for a short period of time might have increased first and then decreased platelets, and there was a dose-response relationship between the cumulative radiation dose and platelets changing.

ABSTRACT

Objective:To predict the therapeutic targets and related signaling pathways of quercetin in the treatment of heart failure (HF) by network pharmacology and molecular docking methods, and further clarify its mechanisms through in vitro cell model. Method:The pharmacological targets of quercetin were obtained by SwissTargetPrediction and Targetnet databases; the heart failure related targets were obtained by Online Mendelian Inheritance in Man(OMIM),GeneCards and Therapeutic Target Database(TTD) databases; the protein-protein interaction(PPI) network was analyzed by STRING database(Search Tool for Recurring Instances of Neighbouring Genes),and the PPI network diagram of quercetin for heart failure target was established. Cytoscape 3.7.2 software was used for analyzing and screening the anti-heart failure network nodes of quercetin, and the obtained targets were enriched with gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis by DAVID database. In order to explore the mechanism of quercetin in the treatment of heart failure,we used cell model to verify the function in heart failure treatment. Results:The predicted results showed that there were 23 targets for the treatment of heart failure, such as Matrix Metallopeptidase-9(MMP-9),androgen receptor(AR),coagulation factor 2(F2),insulin like growth factor 1 receptor(IGF1R),epidermal growth factor receptor(EGFR),janus kinase-2(JAK2),cytochrome P450 family 19 subfamily A member 1(CYP19A1),estrogen receptor-1(ESR1),tumor necrosis factor(TNF),protein tyrosine phosphatase receptor type C(PTPRC) and cytochrome P450 family 17 subfamily A member 1(CYP17A1) etc. The results suggest that quercetin may play a role in the treatment of heart failure by intervening in the physiological processes of cardiovascular cell proliferation and metabolism, regulating hypoxia-inducible factor 1 (HIF-1)signaling pathway and steroid hormone biosynthesis. Conclusion:Quercetin has the characteristics of multi-target,multi-channel and multi-channel in the treatment of heart failure. It may play a role in the treatment of heart failure by regulating MMP-9,EGFR and other key genes, participating in the biological process of cardiac and vascular cell proliferation and metabolism.

ABSTRACT

Objective: To investigate the protective effect and possible mechanism of Diaphragma Juglandis Fructus(DJF) extracts on the rhabdomyolysis-induced acute kidney injury(AKI)in Sprague-Dawley(SD)rats. Methods: Sixty male SD rats were randomly divided into 6 groups: normal control group, model group as well as the four test groups treated with 180 mg/kg and 360 mg/ kg water extract or alcohol extract of DJF. The rats were orally administered once a day by gavage with the designed concentration of test substances or distilled water(for controls)for 30 days. After the last drug administration, all rats except for those in the normal control group were subjected to the intramuscular injection of 50% glycerol to both hind limbs to create the rat model of rhabdomyolysis-induced AKI. The renal function and kidney histopathology were then examined and the serum level of tumor necrosis factor-alpha (TNF-ɑ)and antioxidant indicators were detected at 24 and 48 hours after the glycerol injection. Results: Compared with the normal control group, the urine volume for 24 h as well as the serum glutathione peroxidase(GSH-Px)and total superoxide dismutase(TSOD)activities were significantly decreased, while the serum creatinine(Cr), blood urea nitrogen(BUN), TNF-ɑ and malondialdehyde(MDA)level was significantly increased in the model group(P0.05). However, the intervention with the 360 mg/kg DJF water extract significantly increased the urine volume for 24 h, enhanced the serum T-SOD activity on the 48 h, and decreased the serum Cr, BUN, MDA and TNF-α level on the 48 h(P<0.05 or P<0.01). Conclusions: Both the 360 mg/kg DJF alcohol and water extracts could effectively protect the rhabdomyolysis-induced AKI, which caused by glycerol in SD rats, and the effect of the alcohol extract was better than that of the water extract. The possible mechanism is likely related to the inhibition of inflammation and a positive role in antioxidation.

ABSTRACT

OBJECTIVE: To observe the effect of n-hexane subchronic exposure on the serum level of neuron specific enolase(NSE), neurofilament light chain protein(NF-L) and nerve growth factor(NGF) in rat, and to explore the feasibility of using NSE, NF-L and NGF as biomarkers of n-hexane neurotoxicity. METHODS: Specific pathogen free male SD rats were randomly divided into control group and low-, medium-and high-dose exposure groups, with 6 rats in each group. Rats in the low-, medium-and high-dose exposure groups were given n-hexane solution at doses of 168, 675 and 2 700 mg/kg body mass, respectively, while rats in the control group were gavaged with 0.9% sodium chloride solution, once a day, 5 days a week for 6 weeks. At week 0, 2, 4, and 6 of exposure, the body mass of the rats was weighed, the gait scores were performed, and the serum levels of NSE, NF-L, and NGF were detected.RESULTS: Body mass, gait score and serum levels of NSE and NF-L in rats were statistically significant in terms of the n-hexane exposure dose and exposure time(P<0.01). At the 6 th week of n-hexane exposure, the body mass of the three dose exposure groups was lower than that of the control group(P<0.05), and the gait score was higher than that of the control group(P<0.05). Moreover, the abnormal gait of the rats showed a dose-effect relationship with the increasing n-hexane poisoning dose. At week 2, 4 and 6, the serum levels of NSE and NF-L in these three dose exposure groups were higher than that in the control group(P<0.05). In addition, the serum level of NF-L in rats in the medium-and high-dose exposure groups increased with the n-hexane exposure time increasing and showed a time-effect relationship(P<0.05). The level of serum NGF in rats was statistically significant in the main effects of n-hexane dose and duration of exposure(P<0.05). The serum NGF level in the high-dose exposure group was lower than that in the control group, the low-dose and medium-dose exposure groups(P<0.05). NGF level in serum of rats at week 6 was lower than that at week 0, 2 and 4(P<0.05). CONCLUSION: Both NSE and NF-L in serum can be used as biomarkers for the early effect of n-hexane on peripheral nerve injury. The feasibility of using serum NGF as a biomarker for the early effect of n-hexane on peripheral nerve injury warrants further investigation.

ABSTRACT

Objective@#To investigate the occupational health survey of 1-brominepropane (1-BP) enterprises and understand the impact of 1-BP on the health of occupational exposure population.@*Methods@#The occupational health data of 15 1-BP workers were collected from 3 time nodes in 0 months, June and December, and the effects of occupational exposure to 1-BP on health were analyzed.@*Results@#In the workplace with pure 1-BP, the mean air concentration in the workplace was 26.8 mg/m3, and the personal contact level was 29.7 to 63.4 mg/m3. The occupational health monitoring data showed that white blood cell count (WBC) , red blood cell count (RBC) , aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were compared in 0 months, June, and 12 months, the difference was statistically significant (P<0.05) .@*Conclusion@#During the 12 months observation period, the occupational exposure to 1-BP caused the number of peripheral blood erythrocyte and leukocyte count and the level of alanine transaminase in the workers, but it did not exceed the normal reference range.

ABSTRACT

Objective:To investigate the effect of alteplase on the expressions of Claudin-1 and Claudin-5 proteins in the vascular endothelial cells after thrombolysis in the rats with acute cerebral infarction and its mechanism, and to provide experimental evidence for its clinical application. Methods: The models of acute thrombosis of middle cerebral artery of the rats were established.Fifty-four SD rats were randomly divided into sham operation group, model group and alteplase group (n= 18).The ultrastructure of brain endothelial cells of the rats was observed under transmission electron microscope. The expression levels of Claudin-1 and Claudin-5 proteins in the vascular endothelial cells of the rats were detected by immunoflurorescence and Western blotting methods. Results:The transmission electron microscope results showed that the brain volume in the ischemic area of the rats in model group was significantly increased and the endothelial cells were swollen, some of the cortical and surrounding brain tissues had obvious boundaries, and the thickness of the basement membrane was uniform and the tightly connected structure was very loose and disappeared;the swelling condition of the capillary endothelial cells in infarcted area of the rats in alteplase group was significantly reduced and the thickness of the basement membrane was improved, and most of the tightly connected structures between the brain capillary endothelial cells and the endothelial cells were loose and the fracture was lost and the structure disappeared. The immunofluorescence results showed that the expressions of Claudin-1 and Claudin-5 proteins in the vascular endothelial cells of the rats in alteplase group were significantly improved compared with model group. The Western blotting results showed the expression levels of Claudin-1 and Claudin-5 proteins in the vascular endothelial cells of the rats in model group were significantly decreased compared with sham operation group (P < 0.01 ); compared with model group, the expression levels of Claudin-1 and Claudin-5 proteins in the vascular endothelial cells of the rats in alteplase group at different time points were increased (P < 0.01 ). Conclusion: Alteplase can improve the structure of brain endothelial cells in the rats with acute cerebral infarction, and the mechanism may be related to decreasing the expressions of Claudin-1 and Claudin-5 proteins in the vascular endothelial cells of the rats induced by alteplase.
